Edexcel A CP11: Investigate Photosynthesis Using Isolated Chloroplasts

Abstract:

Understand the light-independent reactions as reduction of carbon dioxide using the products of the light-dependent reactions (carbon fixation in the Calvin cycle, the role of GP, GALP, RuBP and RUBISCO).

Hints & Tips:

- Equipment and solutions should all be kept cold throughout the practical, store solutions in the fridge, and have trays of ice available to store solutions during the practical.

- Phosphate buffer is made from disodium phosphate(4.48 g) and potassium phosphate(1.7 g) made up to 500 mL with distilled water. Keep cold.

-  Sucrose should be made to 0.5moldm-3 and kept cold.

- DCPIP should be used at 0.1%.
